And other stuff like cholesterol.

There are a lot of methods for that.
For example:
Making a coordination complex with the hormones and determining how much complex is formed (with the spectrophotometer).
Or using antibody's from animals for the detection of the pregnancy hormone hCG (agglutination test).

Whether it is possible to do it at home depends on the test you want to do.
