Peptide Synthesizer
Does anybody have any experience or preferences as regards to peptide synthesizers for mid scale preparative work?
Around 100mg or 10- 50 mmol, these are max 10-mer
I've looked at the various ones out there, like ABI the gold standard? AAPPTec, Thuramed, Protien Technologies, CSPS Pharmaceuticals Etc.
I can't seem to decide which may be best.
Working with Rink amide resin, HBTU/ DIEA activation, Fmoc / Alloc and TFA/ TIPS cleavage.
@Sauron, any reccomendations?

As I said in my post : Rink amide MBHA resin from GL Biochem , Shanghai China. Fmoc blocking, removal w/ piperidine and Alloc removal w/ PdTriphenyl
Phosphine/ Phenylsilane
Most interested in ease of use, software interface and automation. Also Only needs to be one at a time but more wouldn't be bad since I need to make

I don't have any experience with the automated synthesizers.
So while I could tell you to go with ABI because it's the gold standard, it really wouldn't address your concerns about interface ease of use etc
would it?
Unfair to the othgers as ABI is the only one I have even studied on paper.
Not that this forum isn't perfect in every way but...have you joined ABRF by any chance? Because they have a forum much more attuned to this sort of
work than SM is.
I joined but don't look in much.
I have one of their white papers, which youmight find of interest. A while ago they decided to assess just how well their member labs were making use
of their peptide synthesizers so they distributed a protocol to make a relatively simple peptide by specific methods using specific reagents etc.
How many of the labs got it right do you reckon?
I think it was less than 50%
I was shocked.

Interesting that you bought from GL Biochem, I got some quotes from them last year for some Boc-protected AAs and some reagents but never bought
anything, because I started to rethink the entire strategy into Fmoc.
Which is why I have a Kg of Boc-Trp sitting here going to waste, but I didn't buy it from GL, but from another CHinese company in Shanghai. Very
inexpensive, like $100.
GL's AA prices seem to be midrange for China, not highest not cheapest.
Except for Alloc with which I have not much familiarity everything else sounds pretty familiar and straightforward.
Piperidine I will make myself as I can't readily buy here without having to justify myself to the drug cops and they often arbitrarily say no anyway.
Silly but that's life.
Didn't they just slap some shipping constraints on HBTU for being energetic?

Nova's great, just expensive.
Have yu thought about HCTU instead of HBTU?
HATU just too costly.
