i hope anyone could help me in extraction and quantifying the polyacetylene in leaves of a plant tissue i did not finde any realiable method for

Starts in the mid 170's. It would appear that you did not search the net well enough as google books has a digital copy with the pages you require.
And this paper should cover your quantification problem.
